祛痰通络汤对早期糖尿病肾病大鼠肾组织TGF-?1表达的影响

- 目的 观察祛痰通络汤对糖尿病肾病(diabetic nephropathies,DN)模型大鼠肾组织转化生长因子-b1(TGF-b1)表达的影响。方法 制作糖尿病肾病大鼠模型。按随机数字表法将大鼠随机分为6组:正常对照组、模型对照组、贝那普利组(1 mg/kg·d-1)、祛痰通络汤低剂量组(3 g/kg·d-1)、祛痰通络汤中剂量组(4.5 g/kg·d-1)、祛痰通络汤高剂量组(6 g/kg·d-1),正常对照组和模型对照组每日均在同一时间予同体积蒸馏水灌胃。各组分别干预8周,采用免疫组化、RT-PCR方法分别检测各组肾组织TGF-b1蛋白和基因水平。结果 肾组织TGF-b1蛋白表达分别为:贝那普利组(1.55±0.12)、祛痰通络汤低剂量组(1.54±0.16)、祛痰通络汤中剂量组(1.49±0.17)、祛痰通络汤高剂量组(1.39±0.25),均低于模型对照组(2.79±0.22),(P均<0.05);肾组织TGF-b1 mRNA的表达分别为贝那普利组(0.35±0.07)、祛痰通络汤低剂量组(0.39±0.03)、祛痰通络汤中剂量组(0.35±0.06)、祛痰通络汤高剂量组(0.32±0.07),均低于模型对照组(0.65±0.09),(P均<0.05)。结论 祛痰通络汤下调TGF-b1在肾组织的表达,可能是祛痰通络汤治疗DN的作用机制之一。
- Abstract:
- Objective To observe the effects of Qutantongluo decoction on the expression of TGF-b1 in renal tissue of diabetic nephropathy(DN) rats, which can help to understand the mechanism of action. Methods The rats models of diabetic nephropathy were established by injecting with streptozotocin intraperi-toneally. All Wistar rats were randomly divided into 6 groups:the normal control group, the model control group, Benazepril Hydrochloridec group(1 mg/kg·d-1), Qutantongluo decoction low dose group(3 g/kg·d-1), Qutantongluo decoction median dose group(4.5 g/kg·d-1), and Qutantongluo decoction high dose group(6 g/kg·d-1). Rats in the normal control group and the model control group were given celiac perfusion of distilled water once at the same time and dose. All the rats were gavaged once daily for 8 weeks.The expression of TGF-b1 at the level of gene and protein in renal tissue were tested by immunohistochemistry and RT-PCR methods. Results Compared with the model control group(2.79±0.22), the expression of TGF-b1 at the level of protein in renal tissue of Benazepril Hydrochloridec group(1.55±0.12)and the Qutantongluo decoction low dose group(1.54±0.16), Qutantongluo decoction median dose group(1.49±0.17), Qutantongluo decoction high dose group(1.39±0.25) decreased significantly(P<0.05). Compared with the model control group (0.35±0.07), the expression of TGF-b1 mRNA in renal tissue of Benazepril Hydrochloridec group(0.35±0.07)and the Qutantongluo decoction low dose group(0.39±0.03), Qutantongluo decoction median dose group(0.35±0.06), Qutantongluo decoction high dose group(0.32±0.07)decreased significantly(P<0.05). Conclusion Qutantongluo decoction can down regulate the expression of TGF-b1 in kidney, which may be one of the mechanisms of Qutantongluo decoction effectiveness in clinical treatment of diabetic nephropathies.
